Platelet Storage at 4¡É and 2¡É : Metabolic, Morphologic and Functional Study in Vitro

Abstract
Platelets stored with agitaion at room temperature for 48 to 72 hours as well as those
kept refrigerate for 24 to 48 hours have been found by different investigators to produce
satisfactory therapeutic effect.
In the Korea Red Cross Blood Center, platelet concentrates are stored at 4¡É for up to
48 hours. However optimum conditions for storage of platelet concentrates have not been
fully defined. So we have examined with metabolic, morphologic and functional changes
following storage. Evaluation was made of PRP stored during 72 hours. During this
period there were not significant changes in platelet count, MPV, pH, plasma glucose
and LDH level both at 4¡É and 22¡É. Platelet aggregation with ADP is better preserved
after 72 hours storage at 4¡É than at 22¡É.
The platelet response to hypotonic shock, its value as an indicator of platelet viability
after storage, showed a more progressive decline with increasing duration of storage at
4¡É than at 22¡É.
After 72 hours storage, the bulky projection and sphericity were prominent with SEM
and platelets were lysed or had a swollen disintegrated internal structure as seen with
TEM both at 4¡É and at 22¡É.
In our studies, significant metabolic and membrane derangements did not occur in
platelets stored either at 4¡É or 22¡É up to 48 hours. So we concluded that the
refrigerated platelets should be transfused at least within 48 hours of collection to
produce a satisfactory clinical response.
